You'll get a lot of info from the users here but it would help if we knew the style you were looking to work in. If it's pixel style then something like Pixen would be great. If you're looking to do hand drawn stuff then photoshop is the way to go. You could even use Google Sketchup to recreate 3d work in a 2d environment.
If I were you, the first thing I'd start with is hand drawn sketches for each frame. You'll cut your work time down dramatically once you get going on the final work.
